The local outfit, which was beaten by Singleton in the dying seconds, has set the standard this time around and handed out a lesson to an understrength Greyhounds 38-4 at McKinnon Field on Friday night.
Ironically, the two sides are scheduled to meet again in the final at Pirtek Park on December 8.
Muswellbrook grabbed the early advantage through Sophie Laing.
However, Tyler Kersley allowed Singleton to draw level at 4-all.
Harmony Liddell and Clare Akauma then touched down, with Akauma also landing a goal, to give the Ramettes a handy 14-4 buffer at the break.
Two further tries to Akauma, as well as a double to Kelsey French, and another to Liddell enabled Muswellbrook to run away with the game.
Shae Pemberton landed a couple of conversions.
In the other clashes, the Merriwa Magpies overran the Aberdeen Tigers 30-4 and the Murrurundi Mavettes pipped the Denman She-Devils 8-6.
