About 8.30am on Saturday, a motorcycle and a utility collided on Putty Road, Howes Valley.
The rider of the bike, a 44-year-old man, sustained serious injuries and was taken by the Westpac Rescue Helicopter Services to John Hunter Hospital.
Tragically, he later died.
The driver of the utility, a 58-year-old man, was uninjured.
He was taken to Singleton Hospital for mandatory blood and urine tests.
Officers from Hunter Valley Police District have now started an inquiry into the incident.
A report will be prepared for the Coroner.
The Howes Valley tragedy comes after another motorcyclist died in a separate incident on Friday night.
Investigators closed the New England Highway's southbound lanes after they were called to Belford shortly before 7.30pm.
Police rendered assistance but the 50-year-old died at the scene.
