Jordan William Thompson, aged 21 months, was rushed to Singleton Base Hospital on Thursday, March 19, 2005, after he was found unresponsive in his home.
A post mortem revealed the toddler had ingested a lethal dose of a prescribed anti-depressant medication.
His death is being treated as suspicious and police hope the reward will encourage someone with information to come forward.
Homicide Squad Commander Detective Superintendent Mick Willing will address the media in Singleton on Monday morning.
