While an investigation is currently underway, police have been told a vehicle was seen parked in Sedgefield cemetery, about 7pm.
It is believed unknown persons then damaged numerous headstones.
In all, 36 gravestones were broken during the incident; having been pushed over with a number of those being smashed.
"Our hearts go out to those people affected," Singleton mayor Sue Moore said in a statement on Saturday.
"We offer our sincere regrets for the distress that this incident has caused in addition to the grief they already suffer.
"I wish to convey the magnitude of my disgust with the perpetrators - and disappointment that they may be people who live among us in our community.
"This is not an attack on a public facility or asset.
"This is an attack on the heart of our community, the people laid to rest in Singleton who are now part of the fabric of our town, and those who love them and remember them.
"I hope the perpetrators are found and held accountable for the damage and, most importantly, the distress they have caused.".
Officers attached to Hunter Valley Police District are now investigating the circumstances surrounding what occurred.
As police continue their inquiries they're asking for anyone who may have witnessed any suspicious activity in the area, or has any information that could assist investigators, to come forward.
It follows after vandals targeted the all-new all abilities playground in Rose Point Park, within days of it being unveiled to the community.
"Significant effort has gone into delivering this particular project to provide facilities for children of all abilities, and it was a project our parks and gardens crew was excited to deliver," Singleton Council's director planning and infrastructure services Mark Ihlein said earlier in the week.
"It is an understatement to say these incidents cut to the heart of our community and the families who benefit from this playground, in particular children with special needs."
